接(jie)種(zhong)(zhong)(zhong)箱(xiang)分(fen)為固(gu)(gu)體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)接(jie)種(zhong)(zhong)(zhong)箱(xiang)和(he)液體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)接(jie)種(zhong)(zhong)(zhong)箱(xiang)兩(liang)(liang)種(zhong)(zhong)(zhong)。固(gu)(gu)體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)接(jie)種(zhong)(zhong)(zhong)箱(xiang)是(shi)一個(ge)用木(mu)料和(he)玻(bo)璃制成(cheng)或由有機玻(bo)璃焊接(jie)而(er)成(cheng)的(de)(de)密閉(bi)小(xiao)箱(xiang)。又分(fen)為雙人(ren)和(he)單人(ren)操作(zuo)箱(xiang)。箱(xiang)體(ti)(ti)可大可小(xiao),一般箱(xiang)體(ti)(ti)長約143cm,寬86cm,總高(gao)154cm,支架76cm。箱(xiang)的(de)(de)上部(bu)左右(you)兩(liang)(liang)側各裝(zhuang)有兩(liang)(liang)扇能(neng)啟(qi)閉(bi)的(de)(de)玻(bo)璃推拉(la)門(men),方便菌(jun)(jun)種(zhong)(zhong)(zhong)進出。窗的(de)(de)下部(bu)分(fen)別設有兩(liang)(liang)個(ge)直徑約13cm的(de)(de)圓(yuan)洞,兩(liang)(liang)洞的(de)(de)中心距(ju)離為52cm(同肩寬),洞口裝(zhuang)有帶松緊帶的(de)(de)袖(xiu)套(tao),以(yi)防(fang)雙手(shou)在箱(xiang)內操作(zuo)時,外界(jie)空氣進入(ru)箱(xiang)內造成(cheng)污染。操作(zuo)時兩(liang)(liang)人(ren)相對(dui)而(er)坐(zuo),雙手(shou)通過袖(xiu)套(tao)伸(shen)入(ru)箱(xiang)內。箱(xiang)兩(liang)(liang)側最好也裝(zhuang)上玻(bo)璃,箱(xiang)頂部(bu)為木(mu)板或玻(bo)璃。箱(xiang)內頂部(bu)裝(zhuang)有紫(zi)外線殺(sha)菌(jun)(jun)燈和(he)照明用日(ri)光燈各一支。箱(xiang)體(ti)(ti)安裝(zhuang)木(mu)板或玻(bo)璃均可,但要注意密封。

液體(ti)菌(jun)(jun)(jun)種(zhong)接種(zhong)箱是專為(wei)移(yi)接液體(ti)菌(jun)(jun)(jun)種(zhong)而(er)設計(ji)的。比(bi)固體(ti)菌(jun)(jun)(jun)種(zhong)箱窄長,單側兩人操作(zuo)。內設軌道(dao)和紫外線燈(deng),箱兩端開有(you)高25cm,寬10cm的長方形(xing)出口,方便菌(jun)(jun)(jun)種(zhong)進(jin)(jin)出,洞口設有(you)小(xiao)推門。進(jin)(jin)出口下處設蒸(zheng)汽源,接種(zhong)時用(yong)蒸(zheng)汽封住(zhu)進(jin)(jin)出口,以防雜菌(jun)(jun)(jun)進(jin)(jin)入箱內。箱背面(mian)設有(you)液體(ti)菌(jun)(jun)(jun)種(zhong)移(yi)接管能(neng)進(jin)(jin)入的小(xiao)孔(kong)。

接(jie)(jie)種箱滅菌(jun)時,用紫(zi)外(wai)線照射30min。如果沒有紫(zi)外(wai)線燈,可用甲醛和(he)(he)高錳酸鉀(甲醛10~14mL/m3+高錳酸鉀5~7g/m3空間)熏蒸30min以上。使用時,先將所(suo)需物品和(he)(he)工(gong)具(ju)放入接(jie)(jie)種箱內,然(ran)后進(jin)行藥劑熏蒸和(he)(he)紫(zi)外(wai)線滅菌(jun),再按無(wu)菌(jun)操作進(jin)行接(jie)(jie)種。接(jie)(jie)種箱的結構簡(jian)單,造價低廉(lian),易消毒滅菌(jun),操作方便,而(er)且人在(zai)箱外(wai)操作,氣(qi)溫(wen)較高時也能作業。缺(que)點是進(jin)出培養基費(fei)工(gong)費(fei)時,每次接(jie)(jie)種前(qian)都(dou)需要(yao)進(jin)行滅菌(jun)。

洗衣機 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
